Chloride toxicity, including its effects on early life stages in fresh and marine waters, remains a major challenge. The consequences of chlorides on aquatic invertebrates is poorly understood as direct effects on physiological mechanisms are hard to establish, and specific target sites are not clearly defined. In contrast, published studies on fishes have been highly successful in pinpointing specific physiological pathways targeted by chloride, which have potential relevance for understanding both the mechanisms of chloride toxicity and the physiological control of chloride homeostasis. A number of mechanisms, including alterations in osmolality, active chloride transport, intracellular amino acid transporters, activation of the cytoskeleton, cell-cell signaling and a number of other pathways are suggested and tested. In this review, we provide a synthesis of this literature and suggest mechanisms that may explain the observed effects of chloride. The relative contribution of each of these pathways to the physiological and toxicological responses to chloride remain to be determined, and our understanding of the most important chloride pathways to target requires further experimental work. Thus, we propose a set of actions to minimize the impact of chloride on freshwater and marine ecosystems. When possible, a combination of both short- and long-term efforts are required, including reducing inputs of inorganic and organic chemicals to wastewater treatment plants (WWTPs), and providing additional effluent flows to restore river ecosystems to pre-treatment levels.Q:
